| Abstract: | Objective: This study evaluates the effect of alpha-tricalcium phosphate cement (U- TCP) and platelet-rich plasma (PRP) on osteogenesis, when used alone or together, comparing to the gold-standard (autologous graft). Material and Methods: Thirty-four Wistar-Kyoto rats were used in this study. A bilateral femur defect was created, and each hole was filled with one of the four types of treatments (autologous bone graft, U-TCP cement, PRP and U-TCP cement+PRP) and evaluated in 4 and 8 weeks. In the control group no filling was applied. The X-ray images provided values of area and longitudinal length of the defect, and the histological images, indicated the new bone formation area. Results: Regarding the effect of the treatments on the bone defect, independently of the time, the X-ray results did not provide sufficient data to confirm important differences in both variables analyzed – area and length (p=0,08). In the histomorphometry, it was observed a better performance of the treatment with autologous bone graft, and it presented significant differences regarding the new bone formation area, independently of the time, compared to the PRP (p=0,05) and control (p=0,041). On the other hand, there were no important differences between autologous graft and the groups U-TCP cement+PRP and U-TCP cement (p>0,05). The treatments PRP, U-TCP cement and U-TCP cement+PRP did not differ significantly from the control group, as well as when they were compared to each other. Conclusion: The data of this study suggest that the treatments with U-TCP cement and PRP, when applied alone or together, do not present positive effect on bone healing.
Objetivo: Avaliar o efeito de cimento de alfa-fosfato tricálcico (U-TCP) e plasma rico em plaquetas (PRP) sobre a osteogênese, quando utilizados isoladamente ou em conjunto, comparando os resultados com o padrão-ouro (enxerto autólogo). Material e Métodos: Trinta e quatro ratos Wistar-Kyoto foram utilizados no estudo. Foi criado um defeito cavitário bilateral no fêmur e cada cavidade foi preenchida com um dos 4 tipos de tratamentos (enxerto autólogo; cimento U-TCP; PRP; cimento U-TCP+PRP), sendo avaliados em 4 e 8 semanas. No grupo controle não foi aplicado nenhum preenchimento. As imagens radiográficas forneceram valores da área e do comprimento longitudinal da lesão, e as imagens histológicas indicaram a área de neoformação óssea. Resultados: Quanto ao efeito dos tratamentos sobre a lesão óssea independentemente do tempo, os resultados radiográficos encontrados não forneceram dados suficientes para comprovar diferenças significativas nas duas variáveis analisadas, área e comprimento (p=0,08). Na histomorfometria, foi observado um melhor desempenho do tratamento com enxerto autólogo, apresentando diferenças significativas quanto à área de neoformação óssea, independente do tempo, em relação aos grupos PRP (p=0,05) e controle (p=0,041). Em contrapartida, não foram verificadas diferenças com significância entre enxerto autólogo e os grupos cimento U-TCP+PRP e cimento U-TCP (p>0,05). Os tratamentos PRP, cimento U-TCP e cimento U-TCP+PRP não diferiram significativamente do grupo controle, assim como não foram evidenciadas diferenças quando comparados entre si. Conclusão: Os dados do presente estudo sugerem que os tratamentos com cimento U-TCP e PRP, aplicados isoladamente ou em conjunto, não demonstram efeito positivo sobre o reparo ósseo. |
