Título: Avaliação da resposta tecidual e da genotoxidade a discos de poli (ácido L/D-láctido) implantados em calota craniana de ratos

Resumo: The aim of present study was to evaluate the local tissue reaction in the skull of rats using the implantation of disks of poly (lactic L/D-acid) and evaluate its genotoxicity, throught the induction of micronuclei in polychromatic erythrocytes (PCE) in the bone marrow of mice. It used 25 male Wistar rats, with 200 and 300 grams. 20 animals underwent surgical procedures and had the discs implanted, and 5 animals received postoperative medication in the same way, serving as a control group for genotoxicity. With a 4. 1 mm trephine drill, two ostectomies circular were done, one in each parietal bone. On the right drilling a disc of poly (lactic L/D-acid ) was inserted, and on the left one it was filled by blood clot (control group). The animals were killed at the appropriate times (15, 30, 90 and 120 days after surgery), by inhalation of isoflurane. In the 20 operated animals the skull was removed and fixed in formalin 10%. The samples were subjected to decalcification and then to paraffinembedded and subjected to cuts of 6 microns in the transverse direction, at the portion of larger diameter discs. The slides obtained were subjected to histochemical technique of staining EH. To evaluate the genotoxicity, both femurs of each animal were removed and the contents of the bone marrow extracted directly on a slide with a drop of fetal calf serum, and mixed. It was made a smear with this material. The slides were dried, fixed in methanol and stained one section of each femur with Giemsa and the other with Acridine Orange. The results were subjected to statistical evaluation by analysis of variance (ANOVA). In histological evaluation, between 15 and 30 days, the bone defect was made clear in both groups, and it is not observed bone formation and resorption episodes or foreign body reaction. In periods of 90 and 120 days in the control group, it was noticed a new formation at the edges of the defect. In the experimental group, there was new bone formation at the edges of the defect and migrating below the site occupied by the disk and the absence of inflammatory infiltrate. Regarding to the evaluation of genotoxicity it was not detected a significant reduction (p <0. 05) in the frequency of EPC in relation to negative control or significant increase (p <0. 05) in the EPC with micronuclei. It can be concluded that the material used in this study is biocompatible and well tolerated by the tissues studied, and found negative for chromosomal mutagenicity when tested.
O objetivo do presente estudo foi avaliar a reação tecidual local, em calota craniana de ratos, frente à implantação de discos de poli (ácido L/D-láctico) e avaliar a sua toxicidade genética, pela indução de micronúcleos em eritrócitos policromáticos (EPC) na medula óssea de ratos. Foram utilizados 25 ratos Wistar, machos, com 200 e 300 gramas. 20 animais sofreram procedimento cirúrgicos e tiveram os discos implantados, e 5 animais receberam medicação pós-operatória da mesma maneira, servindo como grupo controle para genotoxicidade. Com uma broca trefina 4,1mm foram realizadas duas ostectomias circulares, uma em cada osso parietal. Na perfuração direita foi inserido um disco de poli(ácido L/D-láctico), e a do lado esquerdo foi preenchida por coágulo sanguíneo (grupo controle). Os animais foram eutanasiados nos tempos apropriados (15, 30, 90 e 120 dias após a cirurgia), por inalação de isoflurano. Nos 20 animais operados foi retirada a calota craniana e fixada em formalina tamponada a 10%. As amostras foram submetidas à descalcificação e após emblocados em parafina e submetidos a cortes de 6 μm, no sentido transversal, na porção de maior diâmetro dos discos. As lâminas obtidas foram submetidas à técnica histoquímica de coloração HE. Para avaliação da genotoxicidade, ambos os fêmures de cada animal foram removidos e o conteúdo da medula óssea extraído diretamente sobre uma lâmina com uma gota de soro bovino fetal, e homogeinizada. Com este material foi feito um esfregaço. As lâminas foram secas, fixadas em metanol e coradas uma lâmina de cada fêmur com Giemsa e a outra com Acridine Orange. Os resultados obtidos foram submetidos a avaliação estatística pela Análise de Variância (ANOVA). Na avaliação histológica, no período de 15 e 30 dias, em ambos os grupos o defeito ósseo foi bem evidenciado, não sendo observados neoformação óssea, episódios de reabsorção óssea nem reação de corpo estranho. Nos períodos de 90 e 120 dias, no grupo controle, observou-se neoformação nas bordas do defeito. No grupo experimental, houve neoformação óssea nas bordas do defeito e migrando abaixo do local ocupado pelo disco e ausência de infiltrado inflamatório. Quanto a avaliação de genotoxicidade, não foi detectada redução significativa (p < 0,05) da frequência de EPC em relação ao controle negativo nem aumento significante (p < 0,05) nos EPC com micronúcleos. Pode-se concluir que o material utilizado neste estudo é biocompatível e bem tolerado pelos tecidos estudados, sendo considerado negativo para mutagenicidade cromossômica no ensaio realizado.
